Yes and no. Uber does operate in Barcelona but not in the same way as other cities and countries. Uber only works as a taxi app, which means you can order and pay for a ride using the Uber App, but the ride will be in a local Barcelona yellow/black taxi. Instead of having its own Uber drivers, the Uber app for Barcelona allows traditional taxis to use the Uber app at no cost.  Not all Barcelona taxi have chosen to use the app however and Barcelona Taxi organisations continue to protest against Uber. Read more on Uber Barcelona page.
What is the best alternative to Uber in Barcelona? Cabify is perhaps the best Uber alternative in Barcelona. It's similar to Uber and one of the most commonly used alternatives to Uber in Spain. There is also another ride-sharing service called Bolt in Barcelona.
